The study of acute toxicity and antihypoxic activity of new metal complexes of iron

Summary:

The Purpose: to determine the acute toxicity of new metal complexes of iron derived vinylimidazole and to reveal their antihypoxic activity in comparison with Acyzol and other known antihypoxic etomersol and hypoxen. Material and methods. Antihypoxic activity was studied in experiments on nonlinear white mice-males 4 models of acute hypoxia (hypobaric, hypoxia with hypercapnia, hemic, histotoxic). Rates of acute toxicity (LD16, LD LD ) were calculated graphically by the method of Miller and Tainter. Changes in oxidative metabolism in mice under the influence of the most active compounds were assessed by the values of oxygen consumption and rectal temperature. Results. The studied metal complexes of iron are small and moderately toxic. The connection under the code of Tetravim on the severity of antihypoxic effect (the increase in life expectancy male mice compared to control groups on 20-127%) in the range of doses studied (5-250 mg/kg, i/p) is superior to other derivatives of vinylimidazole known and antihypoxic: etomersol (25-100mg/kg, i/p) and hypoxen (50-150 mg/kg, i/p). Conclusion. The obtained data on the pharmacological activity of Tetravim require further clarification of the mechanism of antihypoxic effect and clinical trials with the aim of putting it on the drugs market as a promising antihypoxic agent.

Atorvastatin effect on values of endotheli-al condition and acute inflammation in patients on program hemodialysis

Summary:

Objective: To study the effect of atorvastatin on the values of endothelial condition and acute inflammation in patients on program hemodialysis (PHD). Material and methods: The patients were divided into two groups: 28 PHD patients (main group) received atorvastatin, 20 mg once a day for 30 days, and 26 patients (control group) had PHD only. Endothelin 1-21 (ET) level and von Willebrand factor (vWF) activity were determined by enzyme immunoassay, nitric oxide (NO) levels — spectrophotometrically, C-reactive protein (CRP) — by immunoturbidimetric method, fibrinogen — by Clauss method. Results: After 30 days of observation, control patients were found to have NO and ET increase, as well as the reduced activity of vWF relative to initial data, while fibrinogen and CRP levels scarcely changed. The main group patients had similar changes of endothelial condition values, and their fibrinogen and CRP levels decreased. The correlation between the changes of ET and fibrinogen levels was recorded in the patients of the main group. Conclusion: 30-day administration of atorvastatin, 20 mg per day, lowers CRP and fibrinogen levels in PHD patients. It proves the applicability of statins in this group of patients in case of signs of inflammation.

Summary:

Vibratory disease is one of the leading pathologies in the field of professional diseases, Polymorphism of manifestation is its peculiarity, including disorders of bone remodelling process. The goal of this study was to define peculiarity of bone interchange in patients with vibratory disease of common and local impact. Material and methods. 272 patients took part in the research; they were subjected to determination of mineral density level of bone tissue, markers of osteosis and bone resorption and cytokine profile. Three variants of treatment were used in the period of 12 months. Results. Positive dynamics of ultrasonic densitometry indices, osteogenesis markers (ionized calcium, alkaline phos-phatase), osteoprotogerine in sub-groups with vibrating disease treated with calcium medication and vitamin D was revealed. Conclusion. Research of mineral density of bone tissue and markers of bone remodelling is necessary for prescription a pathogenetic therapy at osteoporosis and osteopenic syndrome in patients with vibrating disease from general to local impact.

Effective monitoring of drug safety in the region

Summary:

The purpose of research is to assess adverse drug reactions, which were registered in Volgograd region, using spontaneous reports. Retrospectively notice cards of suspected adverse drug reactions received by the Volgograd regional center of drugs safety monitoring during 2010 and 2011 were analyzed. Evaluation of reliability of causal relationship «Drug— Adverse Drug Reaction» was carried out in accordance with Naranjo algorithm. In 2011 the volume of registered side effects grew almost 3 times by increasing the activity of doctors than in 2010. We defined the structure of drugs, which caused adverse reactions the most frequently. During these two years doctors rarely reported about adverse drug reactions of type A, so we can suppose that doctors had troubles with detecting causal relationship « Drug —Adverse Drug Reaction».

Comparative evaluation of influence of drugs with antioxidant effect on therapeutic efficiency of radiotherapy and oxidative status in mice

Summary:

The purpose of the study is to carry out a comparative analysis of the effect of melatonin (melaxen) and of 3-hy-droxypyridine (mexidol) on antitumor and antimetastatic influence of radiotherapy and oxidative status in mice with Lewis lung carcinoma. Material: Experiments have been carried out on 95 mice of the line C57BI/6 and weight of 20-22 grams. Cyclophosphan has been intraabdominally administered two times in a dosage of 60mg/kg within the interval of 120 hours — 20-30 min before radiotherapy. It has been located on the area of initial tumor in a dosage of 2 Gr at the same time as cyclophosphan injection. Melaxen and mexidol have been intramuscularly injected in the dosage of 45 and 50 mg/ kg for 14 days. Antitumor and antimetastatic effect of the applied therapy and changes in the oxidative status of the animals have been estimated. The results: Melaxen and mexidol do not decrease antitumour and antimetastatic effects of radiotherapy and prevent the activation of free radical processes in animals with tumors. Mexidol has been more effective than melaxen in correction of superoxide dismutase activity in liver. The drugss under the study do not decrease radiotherapy-induced lipid peroxidation in the initial tumor. Conclusion: Melaxen and mexidol do not decrease the therapeutic efficiency of radiotherapy and correct oxidative status in mice with tumor following antineoplastic treatment.

The research goal is to study the actual problem of self-medication. The Association of the European Self-Medication Industry (AESGP) considers the term «responsible self-medication». The key characteristic is that the patients are responsible for their health. The article presents cases of self-medication thoroughly analyzed in Volgograd Regional Centre of Drug Safety Monitoring. Some of them may be considered as unfavourable and even lethal. Therefore it should be mentioned that there is one more benefit of rational pharmacotherapy with priority of prescriptions made by doctors. Рurposе. To study antimicrobial activity of fume of the essential oil of peppermint against gram-positive and gram-negative bacteria. Materials and methods: The screening study of antimicrobial activity of solutions of essential oil by disk-diffusion method and activity of essential oil fume of own preparation and pharmaceutical form of oil according to standard strains of Staphylococcus aureus, Pseudomonas aeruginosa, Esсherichia coli and 12 clinical strains of staphylococci (6 methicillin-resistant and 6 methicillin-sensitive) has been carried out. Results: Essential oil of own preparation and pharmaceutical form showed equal antimicrobial activity against strains of staphylococci. Essential oil of own preparation has been determined to reveal higher activity against gram-negative strains. Conclusion: Received data have proved the presence of antimicrobial activity against all strains of microorganisms and mostly against staphy-lococci
