Elaborating of a cryoconservation protocol of blood vessels for the tissue-specific matrices

Summary:

Purpose: to create a protocol for complex cryoconservation of blood vessels in polydimethylsiloxane to create tissue-specific matrices. Material and Methods. The material ofthe study was the iliac arteries from postmortem donors, seized in the multi-organ donation, rejected for unclaimed and disposed of later. Polydimethylsiloxane was used as a coolant (PDMS) with a viscosity of 5-25 centistokes (Spectroplast, Russia). Bench testing of cooling and heating ofthe blood vessel in PDMS, testing of heating and cooling of PDMS up to operating temperatures (-80°C) in different ways and variants of its thermal insulation, assessment of different cooling rates ofthe vessel during cryoconservation, modeling of 3 types of working chambers for cryoconservation, sterilization and defrosting of up to 10 vessels, modeling of cryoconservation and thawing of vessels in PDMS under different conditions, selection of substrates from silicone and plastic for uniform fixation of vessels which are of size or size in the working chambers or without them. Results. The developed protocol allows cryopreserved from 1 to 10 or more vessels of all basic sizes both immediately after removal and after transportation to the laboratory, to vary the cooling rate from 1.5 to 300°C/min and above, to be used with different levels of laboratory equipment, as with liquid nitrogen, dry ice, and only with a medical freezer. Conclusion. The developed protocol allows to apply it in cases of complex cryoconservation of blood vessels to create tissue-specific matrices and can be recommended for preclinical testing.

Summary:

Aim: to investigate potential cryo- and radioprotective properties of a polydimethylsiloxane of low viscosity to provide safe and reliable cryopreservation and radiation sterilization of human cadaveric vascular allografts. Material and methods. A consecutive experimental set-up was carried out to establish polydimethylsiloxane (further — PDMS) protective endocellular properties. For this purpose a high-densitymesenchymal stem cells suspension was cryopreserved with 10% DMSO solution; PDMS exocellular protective properties were assessed by cyclic cryoconservation of ^femoral artery segments, while PDMS potential radioprotectiveproperties were assessed by gamma irradiation — induced sterilization of 3 cryopreserved femoral artery segments with gross length about 3cm, and for control grafts a slightly changed protocol with glycerin was used. In this study the dynamic viscosity of all cryopreserved vessels was analyzed with using specially designed experimental device, also electron scanning microscopy with lanthanide cells staining and routine hematoxylin — eosin cell staining were applied. Results. We have demonstrated a prominentexocellular protective effect of PDMS confirmed by a SEM and histological results, and also its mediated radioprotective effect, in particular because of its safer preliminary cryopreservation procedure. Conclusion. Use of PDMS for a cryopreservation and gamma sterilization showed its rationality, however requires further modification of protocols and additional researches.

Summary:

The lists of the diseases connected with radiation effects existing now at the level not only Russia and Belarus, but also at the Union State in general can be enhanced using cumulative scientific data. There are possible variants, as exceptions in the list of separate nosologies, reasons of which are rather caused not by radiation factors, and as introduction of diseases which radiation nature is confirmed by long-term researches. The conducted researches showed that creation of the single list of diseases within the Union State is a really question solving of which will demand not only significant scientific researches, but also serious legislative initiatives.

Summary:

Economic efficiency of various telemedical services are still a subject of scientific discussion. The article considers approaches to a complex estimation of economic benefit and economic efficiency of services in remote education. Calculations are submitted on the basis of realization of remote trainings series on training to receptions of urgent medical aid and occupational medicine for the medical personnel of the large oil-extracting company. Preparation of the program, materials, including videofragments, presentations and demonstrations, and also realization of trainings were carried out by autsourcing by experts of the foreign organizations.

Summary:

Aim: to verify new techniques for human cadaveric vascular allografts cryopreservation, thawing and sterilization for the tissue engineering purposes. We use polydimethylsiloxane (PDMS) as a well-known, promising coolant. This allowed us to completely omit any cryoprotective or vitrifying solutions. Using of PDMS also makes possible an applying these allografts directly after freezing and decellularization and also it will also provide an opportunity to develop secure protocols of tissue— engineered vascular conduits cryopreservation. Matherial and methods. After mathematical modeling of cooling process and its validation the experiment for sealed (isolated) freezing at low temperature conditions of 30 femoral arterial segments has been conducted. The segments were at least 10 cm in length and taken from 15 cadaveric donors in the age of 65-85 years. The freezing process was carried out using the abovementioned coolant— PDMS, and then physico-mechanical properties of these allografts were evaluated with the special Instron machine. According to the results obtained, a modeling of their sterilization conditions was conducted (the grafts were freezed). Results. By physico-mechanical properties validation and restricted histological analysis it was shown that there was an accordance between freezed/thawed allografts properties and native vessels. Conclusion. The abovementioned approach for allografts cryopreservation and thawing was efficient enough for further work in this direction.

Purpose: assessment the heavy psychosomatic and all-somatic cardiovascular and cerebrovascular pathology of patient, transferred an acute I degree radiation sickness, from the general evenly gamma-beta radiation. Conclusions. The subdepressive and disturbing-depressive syndrome of patient, transferred an acute radiation sickness (ARS) of I degree, from the general evenly gamma-beta radiation, was independent risk factor of development of multifocal atherosclerosis; Features of development of all-somatic and psychosomatic pathology of patient are based on a combination of genetic prerequisites, environment influences (the stress caused by accident on the ChNPP) and social factors, influencing on him during a course of life, especially during early socialization. Thus at development of psychosomatic frustration the combination of feature of the mental reaction connected with the personal characteristic and special relationship between mental (stress) and physiological (somatic) by aspects of reaction which led to metabolism violation, to aging, decrease in adaptation opportunities of an organism and development age — dependent pathology took place.

Summary:

Aim: to investigate the dynamics of ulcer surface healing depending on type of radiation ulcer and cell product in murine experimental model of severe local radiation injuries after exposure to X-rays. Material and Methods. Mature Wistar rats were used for experiments (males, weight 180-200 grams). Standard model of severe local radiation injuries was used: X-ray irradiation of animals on a modified RAP100-10 device. Isolation of the stromal-vascular fraction carried out by enzymatic treatment of adipose tissue. Autologous cells transplantation performed on day 20 after irradiation in the case of acute radiation injury model (early radiation ulcers) and day 120 in the case of long-term effects of radiation injury model (chronic radiation ulcers). As a positive control allogeneic MMSC derived from rat bone marrow were used. Results. Application of autologous cell products derived from adipose tissue in animal model of severe local radiation injuries was investigated for the first time. It was shown that usage of stromal vascular fraction of adipose tissue for the treatment of early radiation injuries not only leads to better improvement (as compared with the use of MMSCs), but also pronounced therapeutic effect could be observed at an earlier time. On the model of long-term effects of radiation injuries therapeutic effect was observed for all cell products. Conclusion. Cell-based products derived from adipose tissue are promising material for future research and clinical application for treatment of local radiation injuriesK) Described method allows choosing treatment strategy basing on patient's individual characteristics and the type of local radiation injuries.

Summary:

Aim: to examine the therapeutic efficiency of melanin administered to mice after irradiation at lethal and sublethal doses. Material and methods: Survival and hematological states were studied on CD-1 mice receiving on acute or fractionated whole-body doses on X-rays or gamma-irradiation. Melanin soluble was given with water ad libitum from the first to the 30th-day after irradiation. Results. It was shown that melanin produced a significant therapeutic and protective-therapeutic action against acute radiation injury in the dose range 6,5-7,5 Gy (LD80-LD under our conditions). Cumulative survival melanin treated mice was increased to 14,4%, in control group — 1,9%. After fractionated injury (1 Gy daily, total dose 10 Gy) all mice which consumed melanin remained alive, versus 43,7% in control. Melanin decreased radiation-induced damage and stimulated the hematopoiesis recovery after sublethal exposure (5Gy). Conclusion. The results permit to regard melanin as a therapeutic agent for treatment of radiation injuries.

Summary:

Research objective. The purpose ofthe study was to develop methodology for the formation of groups at risk of developing chronic diseases and complex of measures for their prevention when carrying out a compulsory medical examination of workers. Materials and methods. The results of periodic medical examinations for 2012 were analyzed according to 19 the final acts and reports 61 subjects ofthe Russian Federation. The results ofthe examination ofthe connection between disease and occupation were analyzed according to 50 examination protocols ofthe Burnasyan Federal Medical Biophysical Center of Federal Medical Biological Agency during 200-2010. Results. Based on the results ofthe conducted analysis methodology was developed to assess the risk of occupational disease fully meets the current legislative and regulatory framework of the Russian Federation and enable the individual and group level: to assess the risk of occupational diseases (high, medium, low, no); to identify possible nosological forms of diseases that should be expected, in accordance with the current list of occupational diseases; — to predict the presence or absence of responsibility employer for any case of occupational disease; to generate the necessary recommendations to the employer for the prevention of occupational diseases among workers. Conclusion. The method of formation of risk groups for development of occupational diseases recommended for medical organizations, conducting preliminary and periodic medical examinations.
